ABSTRACT

OBJECTIVETo investigate the function and mechanism of miR-610 in lung cancer cell proliferation and invasion.

METHODSThe expression of miR-610 was detected by real-time PCR in lung cancer cell lines 95D and 95C with different metastatic ability. miR-610 mimics and inhibitor were transfected into 95D and 95C cells, respectively, and CCK-8 kit and BrdU incorporation assay were used to detect the effect of miR-610 on lung cancer cell proliferation. Cell scratch test and Transwell test were used to detect the effect of miR-610 on lung cancer cell invasion. Bioinformatics software was used to predict the potential target genes of miR-610. Dual-luciferase reporter gene system and Western blot were applied to test whether miR-610 regulates GJA3 expression directly.

RESULTSThe expression of miR-610 in the weakly metastatic lung cancer 95C cells was (8.75 ± 0.21) times higher than that in the highly metastatic lung cancer 95D cells (P < 0.01). BrdU incorporation assay showed that the number of proliferating 95C cells was (37.41 ± 2.39)% in the control group and (59.63 ± 4.57)% in the miR-610 mimics transfection group (P < 0.01). The number of proliferating 95D cells was (68.75 ± 4.28)% in the control group and (46.13 ± 3.27)% in the miR-610 mimics transfection group (P < 0.01). Cell scratch test showed that at 24 and 48 hours after scratching, the scratch width of 95C cells in the miR-610 inhibitor group was reduced to (58.74 ± 4.62)% and (34.63 ± 2.73)%, respectively, of the initial scratch width, both were significantly lower than that of blank control group and miR-610 inhibitor control group (P < 0.01 for both). The scratch width of 95D cells in the miR-610 inhibitor group was (88.59 ± 6.92)% and (72.24 ± 5.46)%, respectively, of the initial scratch width, and both were significantly higher than that of the blank control group and miR-610 inhibitor control group (P < 0.01 for both). Transwell assay showed that the numbers of invading 95C cells after transfection with miR-610 inhibitor was (112.4 ± 10.6) cells/square in the miR-610 inhibitor control group and (161.8 ± 12.5) cells/square in the miR-610 inhibitor group, indicating a significantly increased invasion ability of 95C cells in the miR-610 inhibitor transfection group (P < 0.01). The numbers of invading 95D cells after transfection with miR-610 mimics was (178.4 ± 12.3) cells/square in the miR-610 mimics control group and (76.7 ± 5.8) cells/square in the miR-610 mimics group, indicating a significantly decreased invasion ability of 95D cells in the miR-610 mimics transfection group (P < 0.01). These data indicated that miR-610 can suppress the invasion of lung cancer cells.

CONCLUSIONmiR-610 suppresses lung cancer cell proliferation and invasion by targeting GJA3 expression.

ABSTRACT

Objective To study the sedative and analgesic effects of different drugs on patients under-going endoscopic variceal ligation( EVL). Methods Sixty patients undergoing selective EVL were randomly assigned to three groups:pethidine group( A),propofol-fentanyl group( B)and dexmedetomidine-fentanyl group(C). Data concerning blood pressure, heart rate, peripheral oxygen saturation,Ramsay score,endo-scopic operation and anesthesia time, surgeons' satisfaction and adverse reactions were recorded. Follow-up was done 24 hours later to confirm the patients' satisfaction and incidence of intraoperative awareness. Results The Ramsay scores of group B(5. 2 ± 0. 3)and C(3. 5 ± 0. 4)were significantly higher than group A (1. 6 ± 0. 4)(P<0. 05). There was no cough, body movement and high blood pressure during the EVL in group B or C,while the incidences of these symptoms in group A(40%,40% and 30%)were obviously higher (P<0. 05). The surgeons'satisfaction of group B(9. 2 ± 0. 3)and C(9. 6 ± 0. 4)were significantly higher than that of group A(5. 0 ± 0. 4)(P<0. 05). The patients' satisfaction scores of group B(9. 6 ± 0. 3)and C (9. 6 ± 0. 4)were also markedly higher than that of group A(5. 4 ± 0. 3)(P<0. 05). There was no difference of operation time among the three groups. The anesthesia time of group A(44 ± 6)was obviously longer than those of group B(35 ± 4)and C(36 ± 5)(P<0. 05). There was no difference in patients'or surgeons'satis-faction,or anesthesia time between group B and C. But the incidences of bradycardia( 40%),hypotension(30%)and hyoxemia(30%)in group B were obviously higher than those of group C(5%,0,0)(P<0. 05). There was no intraoperative awareness in group B or C. Conclusion Both dexmedetomidine and propofol com-bined with fentanyl are superior to pethidine for patients undergoing EVL,while dexmedetomidine provides bet-ter hemodynamics and respiratory conditions compared with propofol.

ABSTRACT

BACKGROUND: Ischemic osteonecrosis of femoral head can cause dysfunction of the patients and the most frequently advocated treatment for advanced osteonecrosis of femoral head is total hip arthroplasty. But the reason is unknown. A potential cause in patients with osteonecrosis of femoral head is abnormal cancellous bone in the proximal femur which is short of further investigation.OBJECTIVE: To probe into proximal femoral change of patients with ischemic osteonecrosis of the femoral head.DESIGN: A non-randomized concurrent controlled study based on the patients.SETTING: Department of Traumatic Surgery, Affiliated Hospital of Medical College of Qingdao UniversityPARTICIPANTS: This study was done at the Department of Traumatic Surgery, Affiliated Hospital of Medical College of Qingdao University from September 1999 to May 2003. Eighteen patients with osteonecrosis of the femoral head including 12 males and 6 females with the average age of (50 ± 5) years and eighteen patients with osteoarthritis of the hip attended including 10 males and 8 female with the average age of(62 ±3)years were involved in this study.METHODS: Histological examination of the femoral heads and cancellous bone biopsies from four regions of the proximal femur in 18 patients with osteonecrosis of the femoral head following total hip arthroplasty was perforned. Eighteen patients with osteoarthritis were set as control group. All histological specimens were examined in a double-blinded fashion.MAIN OUTCOME MEASURES: The femoral neck, the greater trochanter, the lesser trochanter, the necrosis scoring and necrosis positive rate of shaft of femur of the patients in the two groups.RESULTS: Extensive osteonecrosis was found in the proximal femur up to 4 cm below the lesser trochanter in the group with osteonecrosis. The osteonecrosis scoring was 82 and 16 in the osteonecrosis group and control group respectively and the osteonecrotic positive rate was 63.89% and 19.44% respectively, There was an overall statistically significant difference in the extent of osteonecrosis death in the proximal femur between the two groups ( P < 0. 001 ).CONCLUSION: Bone change of proximal femur might be one of the reasons for the early failure of total hip replacement reported in patients with osteonecrosis of the femoral head.

ABSTRACT

We report a complete genomic sequence of rare isolates (minor genotype) of the SARS-CoV from SARS patients in Guangdong, China, where the first few cases emerged. The most striking discovery from the isolate is an extra 29-nucleotide sequence located at the nucleotide positions between 27,863 and 27,864 (referred to the complete sequence of BJ01) within an overlapped region composed of BGI-PUP5 (BGI-postulated uncharacterized protein 5) and BGI-PUP6 upstream of the N (nucleocapsid) protein. The discovery of this minor genotype, GD-Ins29, suggests a significant genetic event and differentiates it from the previously reported genotype, the dominant form among all sequenced SARS-CoV isolates. A 17-nt segment of this extra sequence is identical to a segment of the same size in two human mRNA sequences that may interfere with viral replication and transcription in the cytosol of the infected cells. It provides a new avenue for the exploration of the virus-host interaction in viral evolution, host pathogenesis, and vaccine development.
